Combined ¹H–¹³C chemical shift perturbations (CSPs) were determined from the corresponding 2D HMQC spectra. To monitor the effect of CDNF binding on the BiP chaperone cycle, we acquired 2D methyl NMR spectra of Ile-labelled full-length wild-type BiP under ATP-regenerating conditions. To investigate whether CDNF binding alters conformational dynamics on the μs–ms timescale, we performed Carr–Purcell–Meiboom–Gill (CPMG) relaxation dispersion experiments on the BiP nucleotide-binding domain (NBD) in the absence and presence of CDNF.
